چکیده

The flocculation tests currently used for the serodiagnosis of syphilis involve the use of an aqueous emulsion of cholesterol, beef heart lecithin, and cardiolipin which undergoes aggregation when treated with Wassermann antibody (1). The r81e of cholesterol in ihis reaction has been depicted as providing a particulate surface which enhances the visibility of the aggregates formed by the antigen-antibody reaction (2). However, the function of the two phospholipides has not yet been clearly defined. Before undertaking further immunological studies on the r61e of lecithin and cardiolipin, an attempt was made to ascertain the purity of these two phospholipides, as prepared customarily for serologic use. Chromatograms of several preparations of beef heart lecithin on Magnesol-Celite indicated the presence of more than one component. The present report describes the isolation of a chromatographically homogeneous, immunologically active lecithin from beef heart lecithin prepared commercially for serologic tests.

A simplified purification of lecithin.

A practical and easily reproducible method for preparing lecithin of a high degree of purity is urgently needed in connection with the use of lecithin in cardiolipin antigens (1, 2). Methods devised for this purpose have been published (3, 4), but the procedures, based on the classic studies of Levene and his coworkers (5, 6), were somewhat tedious and the lecithins so prepared have not always ...

Identification of phosphatidal choline as the major constituent of beef heart lecithin.

In the course of a study of the analytical determination of ester groupings in naturally occurring phospholipides, we found that a specimen of beef heart lecithin, prepared according to the method of Pangborn (l), regularly gave ester values considerably below those obtained with egg lecithin. The preparation was presumably pure, since it was colorless, free of amino nitrogen, had a molar N:P r...

Synthesis of a d- -lecithin, an active antigen component in the serodiagnosis of syphilis.

Although the serodiagnosis of syphilis by various tests has been carried out routinely for many years, our knowledge concerning the chemical nature of the specific antibody and antigen is still too limited to allow a satisfactory interpretation of the serological reactions involved. Valuable contributions towards the elucidation of the structure of several constituents of the antigen have been ...

Evaluation of cardiolipin antigen in the tests for syphilis in leprosy.

Pangborn (14) has shown that an antigen for the serologic diagnosis of syphilis can be prepared by the addition of lecithin and cholesterol to a phospholipid obtained from beef heart. Tests for syphilis using these so-called cardiolipin antigens have given few false positive readings in at least one disease, namely, malaria (7, 8, 15, 16). In leprosy, on the other hand, Rein and his coworkers (...

The cardiolipin-lecithin test for syphilis.

as the antigen for a simplified precipitation test for syphilis. Cardiolipin is the lipid which, in combination with lecithin, gives the alcoholic extract of beef heart its antigenic properties. It is a complex phosphatidic acid with a molecular weight of 2195 and iodine number 120-125. It requires lecithin and cholesterol for antigenicity. It has not yet been synthesized. The cardiolipin-lecit...
